Android normally separates two things: putting the screen to sleep and locking the device. Your screen timeout or auto-lock timer decides when the lock screen actually appears after the display goes dark. The Power button instantly locks option overrides that behavior for the power button specifically.

When this setting is turned on, pressing the power button does both actions at once: the screen sleeps and the device locks immediately, regardless of any longer auto-lock delay you configured. This is useful if you want guaranteed security every time you manually turn off the screen, for example before handing your phone to someone else.

If it is turned off, pressing the power button only sends the screen to sleep. The device then locks after your regular auto-lock timer runs out, which can feel more convenient if you frequently wake the phone again within a few seconds.
